Rybelsus is an oral tablet form of semaglutide, FDA-approved for type 2 diabetes. Unlike injectable GLP-1 drugs, its labeled instructions require it to be taken on an empty stomach with a small amount of plain water, with no other food, drink, or medication for at least 30 minutes afterward.
Why the timing rules exist
Peptide drugs like semaglutide are normally broken down in the digestive tract. Rybelsus is formulated with an absorption enhancer that only works reliably under those specific empty-stomach conditions, which is why the label's timing instructions are strict rather than a general suggestion.
Bioavailability tradeoff
Oral absorption of semaglutide is markedly lower than injectable absorption, which is part of why Rybelsus dosing (up to 14 mg daily) looks numerically larger than injectable Ozempic dosing despite delivering a comparable therapeutic effect at approved doses.
